It took 68 seconds to connect to satellites and show location. Accuracy +-100 to 200 meters. Only one 5floor living house, other houses around much smaller.
No, I am not using a case, only as it is. Yes, I have configured internet connection on my phone, but I use it rarely, so it's switched of all the time. Do you think that could be a problem, some kind of an ineterference?

By so far it means that if I want to have GPS I should go around MIUI and Cyanogenmod?
I try both version 2.2 and 3.1 on my Desire S but no effect, still hard to get satellite and fix (with GPS Status only get 1/1 fix/sats).

When i use custom ROM Virtuous Quattro RC3 on my Desire S GPS work fine. It can get 7/7 Fix/Sats with GPS Status. Anyone know how to get it work on stock HTC Gingerbread? I mean can I get some file in Virtuous Quattro RC3 to be patch to stock ROM?
Explain further. You tried to use this on WHAT ROM's exactly? Read the Notes Section and then the HOW TO POST section above it. Give us good information.
Sounds almost like you have GPS fixing apps installed like Faster Fix or something.
I installed agps.patch.google.build.nossl.v3.1.zip by Recovery on Desire S Android 2.3.5 Kernel 2.6.35.10-ga76c374.
I was once installed GPS Fix but it's uninstalled before using this patch.
I am able to get a lock fast, but after a min or two it looses the lock and I have to exit out. Is there a fix for this "hang up" or can someone point me towards the right direction.

Try GPS Status. Set the app to not allow your screen to lock. Watch it for a few minutes and see if you actually lose signal.
With GPS status is locks and stays locked. I tried re-installing google maps and clearing all the cache/data but still has the same issue.
I've tried CM7, Icecoldsandwich, MIUI and none of those seem to work. They all lose GPS after about 1 min then I have to exit and go back into "navigator" to let it lock on again. Any other thoughts?

This app does installs a customized gps.conf file for your GPS. It has several different profiles to choose from. We recommend to use the default Google profile but it is up to you. Appart from the profiles you can customize the one chosen.
NTP servers
Derek Gordon maintains a special NTP server pool for you. Dispite what everyone thinks, it is not necessarily the one closest to you which is the best. We consider Derek Gordon's server to be the best option to choose because it has the best data and is most frequently updated.

aGPS Alamanacs
Derek Gordon maintains a special aGPS almanac for devices using Qualcomm or similar chipsets. This xtra.bin data file is updated every 30 minutes and is located on a content delivery system maintained by the generous donations of ther userbase.

Donations
To maintain the server we need money. Donations through this app will be divided between Derek Gordon (Crypted) and Zatta on XDA.
Each star costs 2 Euro and will reduce the waiting time before you can install the patch. 0 star users wait 20 seconds, 1 star users 10, 2 star users only 5 and 3 star users and Premium users can install immediately.

Backup and restore
On the first run of this app a backup is made on your SD card. Don't remove it because a backup is only made during the first run (or at an update if you choose to remove it anyway) and no other backup is found on your SD card.
ROM updates
The patch this app provides is located on your /system partition so every time you do update your ROM the patch is overwritten and you will have to redo the patch.
Unless you are using a Cyanogenmod based ROM and your rom has addon.d support. In that case a script will be placed in the addon.d directory to preserve the patch.
It can be disabled in the app's settings but is enabled by default.
Asus users
On ASUS roms a typical file is present at /system/etc/gps, namely the gpsconfig.xml file. This app pathes that file as well, to be specific the PeriodicTimeOutSec parameter. Of course a backup is made of this file as well.
