Question [Q] Backup Stock ROM?

Hi folks. I'm having trouble creating a backup of my stock ROM. Rooted the device (TF101) using "1-Click Transformer Root", using ROM Manager with TWRP recovery. When I choose "Backup Current ROM" from the ROM Manager menu, the device reboots but no file is created.

After reboot I get the standard home screen. Things I've read imply there may be a different mode the device should be booting into, but I don't know that for sure.

Are there some steps to making a ROM backup that I'm missing? Some precursor that needs to be installed before/after/with ROM Manager? Any guidance will be appreciated.
