Do not flash the firmware if you don't know what you are doing!
I.e: flashing a 5.XX.XXX.X firmware over a 3.XX.XXX.X firmware WILL brick your phone ---> QHSUSB_DLOAD

I'm currently running Insert Coin 2.6.0. To install this I would need to flash the latest RUU (Sense 5.5) by running it and then flash this OTA. How do I flash it or install it? Do I need a custom recovery after reverting back to stock with the Sense 5.5 RUU?

No thatll be a pain, flash a nand backup of latest sense 5.5 stock un rooted build then take ota, remember to flash stock recovery as well...

Yes, you can go back to dev edition with the latest dev edition ruu (4.19.1540.9) and you'll be prompted to do the latest ota update.

Dont install a custom recovery after the ruu or you will not be able to apply the ota update.

I already have the RUU and the OTA on my computer but I asked how can I flash the OTA so I won't have to download it after running the RUU.

after you have flashed the ruu, boot the phone normally and copy the ota.zip in your internal storage /obb folder.

then reboot the phone in stock recovery. when you see that image with a red triangle do the volume then power button trick to open the recovery menu (hold volume up and press power then release both button quickly). Select ''apply from phone storage'' then select ''/obb'' and finally select the ota.zip file
