Yes very easy to do. Under menu > settings > display> uncheck the screen rotation box.

For super fast switching I have an app called quick settings. You can make it a shortcut in any number of ways with launcher pro, swype gesture to dock button. Or you can long press search and open the program frm there. If you set it as default long press search then the pop up asking you witch program to use goes away and it just opens.

In quick settings you can cuztimze the visable options. I put orintation at the top of the list. With quick settings shortcuted it becomes super easy and fast to change.

Oh and since I seem to be a rep for quick settings ayway (lol, its free) you can also set ringers and notification to be the same volume. So whe you press the vol buttons on the side of your phone both change.

That always locks it to portrait for me, regardless of whether I'm in landscape or portrait at the time.

I misread your question. There is no way I am aware of to lock in landscape mode, short of opening the keyboard.
