The backlight is the highest consumer of power in the display -- so lowering it would be the best thing.

Otherwise, the LCD draws the least power when all pixels are white (no subpixels active). Power is consumed when a color is displayed. So displaying black activates all three subpixels, so it consumes the most power. This is such a miniscule difference in the grand scheme of things, there are other ways you can conserve power.

There should be a positive effect on AMOLED displays, but that's not the case for the TF300T.
