I am S-on with unlocked boot loader running 4.4.3.
I want to upgrade to lollypop and wanted to know the best way to do that. Can I simply flash the 5.01 ROM in this thread using TWRP or I need to install the update from HTC by locking the bootloader?

Sorry, but I also didn't see a clear explanation there. I gues someone else has to help you.
You need updated firmware to match the title. If you don't have it, the ROM will not work.

If S-On you need to take the OTA Update to update firmware and be able to flash ARHD 34.0.
anyone having this issue? sliding from the side while in full screen mode wouldn't trigger my navigation bar .

The thing is, you can't flash firmware when you're S-Off (correct me if I'm wrong here). You need to revert to stock and then take the OTA.
Off course you can flash when you are s-off, thats the whole idea.
That's completely incorrect. With S-OFF you can flash any firmware. With S-ON you'll need a signed firmware for your specific CID.

What you downloaded is not a RUU but a Nandroid Backup.
If you don't understand any of this, it's best for you to ask in the same thread there so many people will be able to help you.

How to flash:
