Trying to update but fails

I downloaded the new Jelly Bean update from ASUS because I had not yet received it over the air. When I try to flash it using TWRP i immediately get a failed error. I am rooted and the bootloader is unlocked.

1. Do I have to relock the bootloader and if so, how?

2. Can I flash this update using TWRP?

I wiped my cache, Dalvik cache, and System.

Where did you get it from the ASUS site? I can test it out for you. Also unlocked and rooted.

I think those are all for JB 4.1, whereas the new update is 4.2.1.
